From what I have gleened online you should upgrade in the following order. Slot 1 and 2 on the Chalice. Bottom right slot 1 and 2. Bottom left all three. Then it is optional, I will finish the third Chalice slot and then the bottom right and finally the middle bottom.
just get them all before the heroic drops
